Bioluminescence imaging systems have been specially designed for the observation of very low levels of light, resulting for example from chemical or biological reactions. Unlike other types of imaging, the sample is not illuminated but generates the light itself: photo-toxicity is therefore completely absent and acquisition times are long. However, the intensity and dynamics of the observed signals constitute very precise indicators of the chemical or biological reactions taking place within the sample, which is why bioluminescence is frequently used for the measurement and monitoring of calcium flux in samples at both the cellular and organism scales. Observations take place in chambers that are totally opaque to ambient light.
