Wide-field microscopes are dedicated to monitoring in vivo development over the long term in cells, embryos, and fluorescent and non-fluorescent animals. Short exposure times, rapid changes in illumination and an acquisition optimized for the sample depth all limit the photo-degradation of exposed tissues. Some wide-field microscopes are furthermore equipped with temperature and CO2 control systems that ensure the survival of the samples over very long periods. The motorized stages allow to memorize multiple fields of view and to aquire them during the same experiment. These tools aer easy to use and satisfy most needs. Their only limitation is their lack of resolution, which for thick samples in particular can lead to use confocal laser scanning microscopes or spinning disk microscopes.
