Compl\u00e9mentaires des techniques de microscopie confocale classiques, elles offrent un acc\u00e8s privil\u00e9gi\u00e9 \u00e0 l\u2019\u00e9tude des membranes, du cytosquelette, de la mobilit\u00e9 des prot\u00e9ines ou encore aux interactions\u00a0mol\u00e9culaires au sein des \u00e9chantillons vivants.<\/p><p>TRI-Genotoul d\u00e9veloppe ces approches pour r\u00e9pondre \u00e0 des probl\u00e9matiques biologiques vari\u00e9es. La microscopie TIRF (Total Internal Reflection Fluorescence) permet l\u2019observation s\u00e9lective des \u00e9v\u00e9nements se produisant \u00e0 proximit\u00e9 imm\u00e9diate de la membrane plasmique gr\u00e2ce \u00e0 une excitation confin\u00e9e sur quelques dizaines \u00e0 centaines de nanom\u00e8tres. Les techniques de FRAP (Fluorescence Recovery After Photobleaching) permettent quant \u00e0 elles d\u2019\u00e9tudier la mobilit\u00e9 et le renouvellement de mol\u00e9cules fluorescentes dans les cellules vivantes. Les approches FRET (F\u00f6rster Resonance Energy Transfer) et FLIM (Fluorescence Lifetime Imaging Microscopy) donnent acc\u00e8s \u00e0 l\u2019analyse d\u2019interactions mol\u00e9culaires et de proximit\u00e9s nanom\u00e9triques entre prot\u00e9ines marqu\u00e9es par fluorophores, ouvrant la voie \u00e0 une caract\u00e9risation fine des m\u00e9canismes cellulaires dynamiques <em>in situ.<\/em><\/p><p>Les techniques de FRAP, optog\u00e9n\u00e9tique, la photo-activation ou la photo-ablation permettent d\u2019\u00e9tudier et de manipuler le vivant, afin d\u2019analyser la dynamique cellulaire, l\u2019expression g\u00e9n\u00e9tique ou le remodelage de structures cellulaires. Les pinces optiques, bas\u00e9es sur l\u2019utilisation de la lumi\u00e8re, offrent quant \u00e0 elles une manipulation non invasive d\u2019objets cellulaires et permettent de mesurer avec une tr\u00e8s grande pr\u00e9cision des forces et propri\u00e9t\u00e9s physiques \u00e0 l\u2019\u00e9chelle nanom\u00e9trique.<\/p><p>Bas\u00e9es sur l&rsquo;utilisation de la lumi\u00e8re pour capturer et d\u00e9placer des objets, les pinces optiques constituent quant \u00e0 elles une technique non-invasive permettant de manipuler les organites et de mesurer les forces de tension ou d\u2019adh\u00e9sion cellulaires. De plus, le syst\u00e8me d\u2019interf\u00e9rom\u00e9trie localise les objets au nanom\u00e8tre pr\u00e8s pour affiner la mesure de leurs propri\u00e9t\u00e9s
