Les translocations peuvent \u00eatre \u00e0 l\u2019origine de cancers, mais elles peuvent \u00e9galement \u00eatre la cons\u00e9quence de chimioth\u00e9rapies g\u00e9n\u00e9rant ainsi des tumeurs secondaires. La formation des translocations au sein de notre g\u00e9nome reste encore mal comprise, m\u00eame si on sait qu\u2019elle implique le regroupement spatial de deux cassures.<\/h5>\n<p>&nbsp;<\/p>\n<hr \/>\n<figure id=\"attachment_7874\" aria-describedby=\"caption-attachment-7874\" style=\"width: 527px\" class=\"wp-caption alignleft\"><img fetchpriority=\"high\" decoding=\"async\" class=\"wp-image-7874\" src=\"https:\/\/trigenotoul.com\/wp-content\/uploads\/2024\/04\/chromatinD-1.jpg\" alt=\"Chromatin compartment D\" width=\"527\" height=\"466\" \/><figcaption id=\"caption-attachment-7874\" class=\"wp-caption-text\">\u00a9 Arnould et Legube<\/figcaption><\/figure>\n<p style=\"text-align: justify;\">En \u00e9tudiant la r\u00e9paration des cassures double-brin et l\u2019organisation tri-dimensionnelle de notre g\u00e9nome, l\u2019\u00e9quipe de Gaelle Legube au CBI a pu d\u00e9montrer que, suite \u00e0 l\u2019apparition de cassures, notre ADN subit des changements organisationnels de sa structure. En particulier les cassures double brins conduisent \u00e0 la formation d\u2019un nouveau compartiment chromatinien appel\u00e9 compartiment D. Ce compartiment contient diff\u00e9rentes cassures qui se regroupent au m\u00eame endroit, ce qui favorise l\u2019apparition de translocations aux effets n\u00e9fastes pour la stabilit\u00e9 du g\u00e9nome. Cependant, ce compartiment D rassemble \u00e9galement certains g\u00e8nes qui jouent un r\u00f4le primordial dans la signalisation \u00e0 la cellule de la pr\u00e9sence de dommages. La pr\u00e9sence de ces g\u00e8nes dans ce compartiment D faciliterait leur activation, permettant ainsi \u00e0 la cellule de r\u00e9pondre efficacement aux menaces pos\u00e9es par les cassures double-brin de l&rsquo;ADN. Ainsi, la cellule aurait un avantage s\u00e9lectif \u00e0 maintenir ce compartiment D.<\/p>\n<p style=\"text-align: justify;\">Cette \u00e9tude \u00e9claire notre compr\u00e9hension de l\u2019impact de la r\u00e9organisation tri-dimensionnelle de notre ADN sur les m\u00e9canismes que la cellule met en place pour faire face au danger caus\u00e9 par les cassures double-brin de d\u2019ADN. Mais le revers de la m\u00e9daille est que cette r\u00e9organisation peut occasionnellement entrainer des effets n\u00e9fastes pour la cellule en g\u00e9n\u00e9rant des translocations chromosomiques, qui sont des \u00e9v\u00e9nements d\u00e9l\u00e9t\u00e8res \u00e0 l\u2019origine de cancers.<\/p>\n<p>&nbsp;<\/p>\n<blockquote>\n<h5>Publication<\/h5>\n<p><strong>Chromatin compartmentalization regulates the response to DNA damage<\/strong> &#8211; Coline Arnould#, Vincent Rocher#, Florian Saur, Aldo S.
