La plateforme du CMEAB vient de d\u00e9velopper une m\u00e9thode de pr\u00e9paration des \u00e9chantillons par cryofixation permettant de mieux respecter l&rsquo;int\u00e9grit\u00e9 de l&rsquo;\u00e9chantillon.<\/h5>\n<p>&nbsp;<\/p>\n<figure id=\"attachment_5411\" aria-describedby=\"caption-attachment-5411\" style=\"width: 201px\" class=\"wp-caption alignleft\"><img fetchpriority=\"high\" decoding=\"async\" class=\"wp-image-5411\" src=\"https:\/\/trigenotoul.com\/wp-content\/uploads\/2018\/06\/cupule-254x300.jpg\" alt=\"\" width=\"201\" height=\"257\" \/><figcaption id=\"caption-attachment-5411\" class=\"wp-caption-text\">Cupule con\u00e7ue pour l&rsquo;observation en cryo-MEB.<\/figcaption><\/figure>\n<p style=\"text-align: justify;\">La cryofixation des \u00e9chantillons est une technique tr\u00e8s d\u00e9licate, devant \u00eatre utilis\u00e9e dans des conditions optimales afin d\u2019\u00e9viter l\u2019apparition de cristaux d\u2019eau modifiant de fa\u00e7on importante l\u2019ultrastructure des \u00e9chantillons. Pour se faire, La cryofixation \u00e0 haute pression est actuellement la seule technique permettant de cryofixer les \u00e9chantillons sans utilisation de surcouches de protection (cryoprotectant) dont les \u00e9paisseurs, avoisinant les 200\u03bcm, alt\u00e8re l&rsquo;aspect de ce dernier.<\/p>\n<p style=\"text-align: justify;\">La plateforme du CMEAB a pour cette raison choisi la cryofixation haute pression comme technique privil\u00e9gi\u00e9e afin de pr\u00e9parer des \u00e9chantillons pour l\u2019observation apr\u00e8s cryofracture en MEB. Le verrou technologique rencontr\u00e9 \u00e9tait l\u2019absence d\u2019adaptateur permettant la fixation des cupules du cryofixateur haute pression.
