Des chercheurs du LAAS (CNRS), en partenariat avec les plateformes de TRI-Genotoul du CPTP et de l&rsquo;Restore, sont parvenus, gr\u00e2ce au gravage laser r\u00e9alis\u00e9s \u00e0 l&rsquo;aide de microscopes multiphotons, \u00e0 fabriquer des \u00e9chafaudages 3D autoportants. Ces supports ont \u00e9t\u00e9 ensuite\u00a0 ensemenc\u00e9s des cellules neurales puis caract\u00e9ris\u00e9s \u00e0 l&rsquo;aide d&rsquo;approches avanc\u00e9es d&rsquo;imagerie par fluorescence 3D. Les r\u00e9sultats publi\u00e9s dans la revue <strong>Small<\/strong> de Advanced Science news se r\u00e9v\u00e8lent particuli\u00e8rement encourageants.<\/h5>\n<p>&nbsp;<\/p>\n<figure id=\"attachment_4920\" aria-describedby=\"caption-attachment-4920\" style=\"width: 908px\" class=\"wp-caption alignleft\"><img fetchpriority=\"high\" decoding=\"async\" class=\"wp-image-4920 size-full\" src=\"https:\/\/trigenotoul.com\/wp-content\/uploads\/2017\/06\/publiaccardo.jpg\" alt=\"Matrice cr\u00e9\u00e9e par les chercheurs\" width=\"908\" height=\"271\" \/><figcaption id=\"caption-attachment-4920\" class=\"wp-caption-text\">Matrice cr\u00e9\u00e9e par les chercheurs<\/figcaption><\/figure>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<hr \/>\n<p style=\"text-align: justify;\">La technique ici employ\u00e9e, autorise un processus de fabrication beaucoup plus fin (\u2248 200 nm) que les technologies habituelles d&rsquo;impression 3D classiques bas\u00e9es sur la mod\u00e9lisation de d\u00e9p\u00f4ts fusionn\u00e9s, l&rsquo;impression jet d&rsquo;encre, le s\u00e9chage s\u00e9lectif au laser ou la technologie \u00ab\u00a0polyjet\u00a0\u00bb.<\/p>\n<p style=\"text-align: justify;\">Les \u00e9chantillons ainsi cr\u00e9\u00e9s ont ensuite \u00e9t\u00e9 \u00e9tudi\u00e9s par de la microscopie \u00e9lectronique \u00e0 balayage (SEM) pr\u00e9cisant ainsi la morphologie des cellules et les connexions cellulaires autour de l&rsquo;architecture 3D. Le positionnement du noyau, inaccessible par l&rsquo;imagerie SEM conventionnelle, est quant \u00e0 lui d\u00e9voil\u00e9 par microscopie de fluorescence en feuille de lumi\u00e8re et imagerie confocale multiphotonique. Ces diff\u00e9rentes techniques ont mis en \u00e9vidence de longues extensions neuritiques et une colonisation cellulaire optimale autour et \u00e0 l&rsquo;int\u00e9rieur de l&rsquo;\u00e9chafaudage 3D.
