Negash\" width=\"450\" height=\"218\" align=\"right\" \/>Afin de s\u2019affranchir des limites de r\u00e9solution impos\u00e9es par les <a href=\"#loisoptiques\">lois de l\u2019optique*<\/a> et ainsi atteindre de plus hautes r\u00e9solutions, diff\u00e9rentes technologies de microscopie de fluorescence ont \u00e9t\u00e9 r\u00e9cemment d\u00e9velopp\u00e9es. Regroup\u00e9es sous les termes de microscopie de \u00ab super-r\u00e9solution \u00bb ou \u00ab nanoscopie \u00bb, elles permettent de localiser, in vivo, au sein m\u00eame des cellules, des compos\u00e9s \u00e0 l\u2019\u00e9chelle de la mol\u00e9cule unique (r\u00e9solutions lat\u00e9rales de l\u2019ordre de 10 \u00e0 90 nm). Ces technologies qui font soit appel \u00e0 des principes semblables au pointillisme (PALM\/STORM) soit \u00e0 de l\u2019illumination structur\u00e9e (SIM) ou \u00e0 de la d\u00e9pl\u00e9tion laser (STED) ont valu \u00e0 leurs auteurs, E. Betzig, S. Hell et W. Moerner, l\u2019attribution du prix Nobel de Chimie en 2014.<\/h5>\n<p>&nbsp;<\/p>\n<blockquote>\n<h5 style=\"text-align: justify;\">Mener une recherche comp\u00e9titive dans de nombreux domaines de la biologie requiert dor\u00e9navant l&rsquo;acc\u00e8s \u00e0 la microscopie super-r\u00e9solue, raison pour laquelle TRI d\u00e9veloppe un p\u00f4le de microscopie de super-r\u00e9solution b\u00e9n\u00e9ficiant de ces diff\u00e9rentes approches et ouvert \u00e0 l\u2019ensemble de la communaut\u00e9 scientifique.<\/h5>\n<\/blockquote>\n<p>&nbsp;<\/p>\n<h3 style=\"text-align: justify;\">\u00a0\u00c9quipements Industriels<\/h3>\n<hr \/>\n<ul style=\"list-style-type: square;\">\n<li style=\"text-align: justify;\">\u00c9quipements de type dSTORM\u00a0 et STED au CPTP. Le premier d\u00e9veloppe des approches de type dSTORM monocouleur\u00a0 en microscopie TIRF, avec un laser 640 nm de 400 mW. Quand au second, le STED, son originalit\u00e9 r\u00e9side dans le fait qu&rsquo;il est le seul microscope de super-r\u00e9solution permettant de travailler sur coupe de tissus en 3D, permettant ainsi d\u2019acc\u00e9der \u00e0 la physiopathologie humaine avec une r\u00e9solution tr\u00e8s fine (30 nm).<\/li>\n<li style=\"text-align: justify;\">Un \u00e9quipement similaire, le N-Storm est op\u00e9rationnel au sein des plateformes r\u00e9unies sous la banni\u00e8re commune LICT (CBD \/ CBI). Il utilise les m\u00eames technologies que les pr\u00e9c\u00e9dents mais permets des observations dans des longueurs d\u2019ondes plus vari\u00e9es.<\/li>\n<li style=\"text-align: justify;\">Un \u00e9quipement de type ELYRA PS1 de la soci\u00e9t\u00e9 Zeiss a \u00e9t\u00e9 implant\u00e9 sur le site de l\u2019I2MC. Ce microscope de super-r\u00e9solution utilise un concept d\u2019analyse des nano-interactions au niveau des contacts entre diff\u00e9rents types cellulaires ou entre les cellules et la matrice extracellulaire. Gr\u00e2ce \u00e0 cet \u00e9quipement, il est possible d\u2019analyser un m\u00eame \u00e9chantillon par les trois des technologies cit\u00e9es pr\u00e9c\u00e9demment, SIM et STORM ou PALM, afin d\u2019obtenir un r\u00e9sultat extr\u00eamement fin, permettant de localiser avec pr\u00e9cisions des mol\u00e9cules uniques.<\/li>\n<\/ul>\n<p>&nbsp;<\/p>\n<h3 style=\"text-align: justify;\">Technologie de R&amp;D<\/h3>\n<hr \/>\n<p style=\"text-align: justify;\">Deux microscopes en\u00a0 d\u00e9veloppement, bas\u00e9s sur une technique couplant l&rsquo;Illumination structur\u00e9e SIM et de localisation 3D, sont op\u00e9rationnels au sein du site LITC. Int\u00e9grant les derni\u00e8res innovations en imagerie par illumination structur\u00e9e provenant des laboratoires de microscopie de renomm\u00e9e internationale (IPHT &#8211; Allemagne, Institut Fresnel &#8211; France, ISMO &#8211; France, Jenelia Lab &#8211; USA), ils permettent de s&rsquo;affranchir des contraintes exp\u00e9rimentales rencontr\u00e9es dans les \u00e9quipements classiques de super-r\u00e9solution. Bas : Exemple d&rsquo;imagerie conventionnelle et super-r\u00e9solue \u00e0 80nm de r\u00e9solution d\u2019adh\u00e9sion focale (\u00e9chelle 640nm).<\/figcaption><\/figure>\n<p style=\"text-align: justify;\"><strong>\u00a0Ces \u00e9quipements pourront \u00eatre coupl\u00e9s \u00e0 des techniques existantes telles que de l&rsquo;opto-manipulation :<\/strong><\/p>\n<ul>\n<li style=\"text-align: justify;\">\u00a0SIM conventionnel TIRF 2D et SIM 3D \u00e0 une fr\u00e9quence temporelle de 20 images \/ s. Cette technique autorisera ainsi l&rsquo;imagerie super-r\u00e9solue\u00a0 de ph\u00e9nom\u00e8ne rapide telle que la mitose et l&rsquo;endocytose,<\/li>\n<li style=\"text-align: justify;\">SIM non-lin\u00e9aire 2D rapide autorisant une r\u00e9solution de 50nm \u00e0 l&rsquo;aide de mol\u00e9cules photo-activables en GFP (fr\u00e9quence d&rsquo;acquisition : 5 images \/ s). Elle sera d\u00e9di\u00e9e \u00e0 l&rsquo;imagerie du cytosquellete et de la chromatine (accessible au printemps 2016),<\/li>\n<li style=\"text-align: justify;\">Imagerie 3D SIM isotrope mutilicouleurs \u00e0 un seul objectif afin de d\u00e9mocratiser l&rsquo;imagerie isotropique super-r\u00e9solue en utilisant un statif de microscope conventionnel (accessible en fin d&rsquo;ann\u00e9e 2016),<\/li>\n<li style=\"text-align: justify;\">Imagerie SIM TIRF 2D associ\u00e9e \u00e0 la localisation 3D par collection supercritique, en collaboration avec AbbeLight (impl\u00e9mentation en juillet 2016).
