En outre, la plateforme de l&rsquo;IBCG c&rsquo;est quant \u00e0 elle doter d&rsquo;un syst\u00e8me similaire nSTORM.<\/h5>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">La microscopie confocale est une approche approximative de la localisation des plus petits organites cellulaires, des domaines de sp\u00e9cialisation intracellulaires, ou bien encore des prot\u00e9ines membranaires. Le d\u00e9veloppement r\u00e9cent de microscopes de super-r\u00e9solution, ou nano-microscopes, permet de d\u00e9passer la barri\u00e8re de diffraction optique qui limite la r\u00e9solution des approches conventionnelles, rendant ainsi possible la visualisation d&rsquo;objets de l&rsquo;ordre de quelques dizaines de nanom\u00e8tres.<\/p>\n<figure id=\"attachment_3193\" aria-describedby=\"caption-attachment-3193\" style=\"width: 524px\" class=\"wp-caption alignright\"><img fetchpriority=\"high\" decoding=\"async\" class=\"wp-image-3193\" title=\"R. Houmadi (plateau imagerie CPTP)<\/figcaption><\/figure>\n<p style=\"text-align: justify;\">L&rsquo; approche de super-r\u00e9solution\u00a0 la plus pr\u00e9cise (r\u00e9solution de 20 nm), est celle de la microscopie de localisation. Elle repose sur le principe de la d\u00e9tection d\u2019une mol\u00e9cule unique et de la localisation du centre de sa tache de diffraction. L\u2019accumulation des acquisitions permet alors la reconstitution d\u2019une image super-r\u00e9solue. Le PALM (PhotoActivated Localization Microscopy), et le STORM (STochastic Optical Reconstruction Microscopy) sont autant de techniques de reconstruction d&rsquo;image par pointillisme permettant d\u2019isoler les mol\u00e9cules uniques \u00e9mettrices de fa\u00e7ons diff\u00e9rentes.<\/p>\n<p style=\"text-align: justify;\">Le dSTORM et nSTORM ouvrent ainsi un nouveau champ d&rsquo;exploration pour interroger les d\u00e9tails de la localisation des mol\u00e9cules r\u00e9gissant les interactions cellulaires \u00e0 une r\u00e9solution de quelques dizaines de nanom\u00e8tres sur pr\u00e9paration fix\u00e9e.<\/p>\n<hr \/>\n<p style=\"text-align: justify;\"><strong>Les plateaux techniques d&rsquo;<a title=\"OUR PLATFORMS\" href=\"https:\/\/trigenotoul.com\/en\/tri-genotoul\/our-platforms\/\">imagerie cellulaire du CPTP<\/a> et\u00a0 de <a title=\"OUR PLATFORMS\" href=\"https:\/\/trigenotoul.com\/en\/tri-genotoul\/our-platforms\/\">l&rsquo;IBCG<\/a> ont acquis une v\u00e9ritable expertise en d\u00e9veloppant des approches de type STORM\u00a0 en microscopie TIRF.
