{"id":2855,"date":"2015-04-08T13:24:52","date_gmt":"2015-04-08T11:24:52","guid":{"rendered":"http:\/\/trigenotoul.com\/?p=2855"},"modified":"2015-04-08T13:24:52","modified_gmt":"2015-04-08T11:24:52","slug":"confocal-video-microscopy-and-laser-nanosurgery","status":"publish","type":"post","link":"https:\/\/trigenotoul.com\/en\/video-microscopie-confocale-et-nano-chirurgie-laser\/","title":{"rendered":"CONFOCAL VIDEO-MICROSCOPY AND LASER NANO-SURGERY"},"content":{"rendered":"<h5 style=\"text-align: justify;\"><a href=\"https:\/\/trigenotoul.com\/wp-content\/uploads\/2014\/07\/CBI.jpg\"><img decoding=\"async\" class=\"alignleft size-full wp-image-608\" src=\"https:\/\/trigenotoul.com\/wp-content\/uploads\/2014\/07\/CBI.jpg\" alt=\"CBI\" width=\"105\" height=\"77\" \/><\/a> Le couplage de l\u2019imagerie confocale aux techniques de micromanipulation du vivant autorise une approche multi-\u00e9chelle de l\u2019\u00e9tude de l<em>&lsquo;<\/em>ensemble des lois qui d\u00e9terminent la forme et la structure de tissus, la <em>morphog\u00e9n\u00e8se. <\/em>L\u2019\u00e9tude des forces localement exerc\u00e9es par une cellule peut \u00eatre alors int\u00e9gr\u00e9e et analys\u00e9e en relation avec la dynamique des cellules composant l\u2019ensemble du tissu, permettant d&rsquo;\u00e9laborer un mod\u00e8le biophysique multi-\u00e9chelle de cette morphog\u00e9n\u00e8se&#8230; C&rsquo;est le d\u00e9veloppement de ces nouvelles techniques qui a aujourd&rsquo;hui permis de d\u00e9couvrir que lorsque une cellule rentre en <em>apoptose<\/em>\u00a0 ou mort cellulaire, elle d\u00e9forme localement le tissus auquel elle est int\u00e9gr\u00e9.<\/h5>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">Le LBCMP a mis en place sur le site TRI \/ CBI une technique de nano-chirurgie laser (Ropper Scientific\/Leica) associ\u00e9e \u00e0 l\u2019analyse d\u2019image de tissus de drosophile en 4D. La nano-chirurgie s\u2019effectue \u00e0 l\u2019\u00e9chelle subcellulaire \u00e0 l\u2019aide d\u2019un faisceau laser impulsionnel. Elle permet par exemple de sectionner de mani\u00e8re extr\u00eamement pr\u00e9cise, dans un tissu et sans induire d\u2019\u00e9chauffement important en raison de la tr\u00e8s br\u00e8ve dur\u00e9e de l\u2019impulsion laser, des filaments du cytosquelette cellulaire ou des jonctions entre cellules. Gr\u00e2ce \u00e0 cette technique, l\u2019\u00e9quipe de Magali Suzanne (CBI) a r\u00e9v\u00e9l\u00e9 le surprenant comportement des cellules <em>apoptotiques<\/em> (en phase de mort cellulaire) [1]. Figure modifi\u00e9e d&rsquo;apr\u00e8s publication [1]<\/figcaption><\/figure>\n<p style=\"text-align: justify;\">Dans ce processus, l\u2019analyse d\u2019images et de nano-chirurgie laser ont permis de quantifier m\u00e9caniquement et morphologiquement les cellules voisines en l\u2019absence ou en pr\u00e9sence de tension apoptotique. La relation entre la morphologie des cellules formant les plis et leurs tensions de surface mesur\u00e9es par nano-chirurgie (fig. 2) a ainsi pu \u00eatre v\u00e9rifi\u00e9e.<\/p>\n<p style=\"text-align: justify;\">Ces donn\u00e9es ont permis d\u2019appuyer l\u2019id\u00e9e que la force produite par la cellule apoptotique peut induire l\u2019augmentation des forces de tensions superficielles au niveau du sommet des cellules voisines, formant ainsi les futurs plis.
